is it a standard 2jz with standard ecu and loom or aftermarket?
ide be checking
idle control valve
map sensor
crank and cam sensors...
in that order
when you say running on approx 4 cyl how are you determining this? low idle/almost stalling?
Lately when I start the engine, it runs on approx 4 cylinders, belts out lots of black smoke until I give it a couple of good revs then all is well. I have changed the plugs - still the same.
Another interesting thing is that if the engine does not fire 1st go, it takes a lot of cranking the second time to fire it, sometimes to the point of nearly flattening the battery.

i would start by checking the injectors. the may be leaking residual pressure from their last cycle and in return making it run on four on startup due to being flooded.
