I had the same problem with my Mark II, except that it was the factory heater line that was the highest but I found it wont be a problem as long as you bleed it properly. I did it by sitting a coke bottle in the top of the radiator and cutting the bottom off. Fill it up enough so that it becomes the highest point, that and parking on an inclined driveway helped.
This should be fine without any other reserviors as long as there is no leaks in the system to allow air in (such as a cracked hose).
The only other problem is removing the bottle and replacing the cap after you are done.
As for filling a 1UZ, I wouldnt have a clue but there shouldnt really be a reason why you cant fill it from the radiator. Also, what do you mean about the inlet/outlet location - they on the same tank or something? As for draining, doesnt your radiator have a drain plug, or are you referring to drain points on the engine?
