I've just read this and wondered what the Hell "Full House" and those really irritating Olsen twins had to do with putting a 1UZ into an RT141. Then it dawned on me with the help of a beer or two that this has been done but not with a 1UZ to my knowledge but no reason why it can't. Why I say it's been done was "Back in the day" a mate of mine had one with a 289 Windsor in it. Another mate had a RWD Magna wagon with a P76 V8 in it and the same guy that had the Corona also had a Mark one Escort with a Rover V8 in it (This is the early '90s in NZ where 1UZs were not cheap but we all wanted one). At the time I had the SL/R5000 Torrie (dumb mistake selling that) and a very worked Mini for a daily (I regret selling that even more than the Torrie). So it can be done, I don't recall any issues with chassis flex but remember the UZ family of engines are a 90 degree V so the strut towers may cause an issue with clearances.
